Products / Services
EnVivo's lead product is an orally bioavailable 7-nAChR agonist (EVP-6124) being tested in Phase IIa clinical trials for treatment of Alzheimer's Disease and schizophrenia. The company is also developing several preclinical programs, including an epigenetics program based on Histone Deacetylase inhibition (HDACi) for cognition in Alzheimer's disease and programs investigating the uses of Gamma Secretase Modulator (GSM) and PDE10 inhibitor.
Technology / Differentiation
Acetylcholine (ACh) is a brain neurotransmitter involved in cognition. Selective activation of an alpha-7 nicotinic acetylcholine receptor (7-nAChR) may alleviate the cholinergic deficit present in Alzheimer's Disease without the undesirable side-effects caused by overactivation of other nAChRs or muscarinic acetylcholine receptors involved in addiction, cardiovascular or gastrointestinal functions. Histones and DNA are integrated components of chromatin, whose quaternary structure plays a primary role in which genes are transcribed or repressed. The acetylation state of the histones can impact the probability of gene transcription; Histone Deacetylases (HDACs) are families of enzymes critical for regulating the aceylation state in the genome. Gamma-secretase modulation reduces production of the peptides contributing to the amyloid plaques associated with Alzheimer's Disease. EnVivo applies a proprietary and novel strategy to CNS drug discovery. Using well-validated models of specific CNS disorders, the company conducts high throughput screening in intact organisms to discover new small-molecule therapeutics. Since the screen detects improvements in disease symptoms, it does not necessarily rely on prior understanding of the disease mechanism. EnVivo's technology can screen for drugs acting at multiple cellular and molecular sites within the complex neural circuits that regulate behavior, making it well-suited for CNS applications.

More than four million Americans currently have Alzheimer's disease. As life-span increases, a projected 11-16 million Americans will develop Alzheimer's by 2050. Alzheimer's is now the third most expensive disease to treat in the U.S., costing close to $100 billion annually. Schizophrenia is a psychiatric condition affecting approximately 4.6 billion individuals worldwide. Its market is expected to grow steadily and reach $4.3 billion in 2011. CNS disorders collectively account for more hospitalizations, long-term care and chronic suffering than nearly all other diseases combined.
Status
EnVivo Pharmaceuticals announced its agreement in April 2009 with Mitsubishi Tanabe Pharma Corporation (MTPC) for the development and commercialization of its lead product candidate, EVP-6124, in Japan and several other Asian countries.
