| Linaclotide is an agonist of guanylate cyclase type-C, a receptor found on the lining of the intestine. In preclinical testing, linaclotide was shown to decrease visceral pain, increase fluid secretion into the intestine, and accelerate intestinal transit. Linaclotide is an orally administered therapeutic designed to exert its effects on the intestine with minimal systemic exposure. |

According to the Mayo Clinic, as many as one in five American adults currently have IBS. This condition accounts for 12% of adult visits to primary care physicians and is the most common disorder diagnosed by gastroenterologists, which translate into health care costs exceeding $25 billion annually. Chronic constipation afflicts as many as 26 million Americans; symptoms include: hard and lumpy stools, straining during defecation, a sensation of incomplete evacuation and fewer than three bowel movements per week.

| In October 2008, Ironwood presented the results of its Phase 2b study assessing linaclotide's safety and efficacy in 420 patients with IBS-C. Analysis of the data indicated that once-daily oral dosing of linaclotide, across a range of doses, significantly reduced abdominal pain and improved constipation symptoms in patients with IBS-C throughout the 12-week period. These results support the advancement of the company's novel compound into Phase 3 clinical trials. Ironwood and Forest Laboratories have a 50/50 partnership in the United States to co-develop and co-market linaclotide; the former company retains exclusive rights to linaclotide outside of North America. Ironwood has raised $281 million in private equity financing to date. |
